Supplemental Data Involvement of MicroRNA in AU-Rich Element-Mediated mRNA Instability

چکیده

Search of the Genes that Are Required for ARE-Mediated mRNA Decay in Drosophila S2 Cells Double-stranded RNAi was used to evaluate a number of different genes in ARE-mediated RNA stability in S2 cells. The RNAi treatments were performed as described in the Experimental Procedure. RT-PCR was used to confirm the knockdown of corresponding mRNA. Real-time PCR was used to measure the stability of ARERNA. The results are summarized in Table S1. It should be noted that observing no change in the screen of a given gene cannot exclude its involvement in ARE-mediated mRNA stability because there could be many explanations such as the protein of this gene may still exist at a low level that is still sufficient for its function in ARE-RNA degradation or other gene(s) may compensate for the function of this gene.
